Off the top of my head...
Battletech- One player steers the Mech, the other player controls the upper body for attacking. It's fun as hell.
Streets of Rage series- Beating up generic bad guys en masse, with a two-player specific move or two to boot.
TMNT: THe Hyperstone Heist- Easy as hell, but fun.
Gunstar Heroes- Shit is flying and blowing up everywhere. Fast paced and fun.
World Series Baseball '95- Arguably the best baseball game on the Genesis.
NBA Jam T.E.- Arcade basketball, two on two, fast paced and over the top... it's an enjoyable game with two players.
Master of Monsters- If you've got a friend up likes turn-based RPGish games, this game's a great way to blow about two hours.
Herzog Zwei- RTS and shmup combine to make for a great way to go head to head via split screen.
Soldier of Fortune- Overhead run 'n gun that can get bit annoying if one player hangs back too often, or is a greedy shit with collecting the dropped money, but it can be enjoyable.
Bio-Hazard Battle- Two player shmup taking on bugs. What more do you want?
SFII:SCE/SSFII- It's Street Fighter, it's solid fighting, it's a no brainer really.

Forgot about Air Buster. A solid, two-player shmup.
Wait, Street Fighter's cooperative? You mean two people can play as Ken and Ryu at once and double team poor little Chun Li? That's just evil.

World of Illusion. With two capes taking up so much screen this game becomes a mite messy and thankfully harder.

Gunstar Heroes. Similar to WoI except that it's messy even on 1P.

That's what comes to mind right now. The Genesis has a terrific lineup of co-op games. Arguably the best of any console.
It helps the co-op process along when one player gets on the other player's nerves ;)
And for those wondering, Master of Monsters can be played 1 human to 3 CPU, 2 on 2, 3 on 1, and all human. So while you may end up with a versus match, you can still team up on the CPU players.

Lost Vikings is great on three player (2-player kinda' sucks though)
Second Samurai is quite funny and over the top
Monster Lair is alright (nothing special)
I got some decent time out of Dinosaurs for Hire (nothing special, but I find it entertaining)
Two Crude Dudes is a whole lot of fun

The first Toejam and Earl. A 2 player Roguelike where you can high five eachother to trade health? Awesome.
Seriously, if you are easily entertained, Pit-Fighter co-op is hilarious. The Genesis version is at least playable too.
Nothing better than throwing motorcycles at biker chicks.
I have to mention Soul Star as one of the most memorable and original co-op experiences.
Sorry it's not strictly a Genesis game.
Gain ground is an awesome 2 player game
Gunstar Heroes, Streets of Rage 2, World of Illusion Starring Mickey Mouse and Donald Duck and ToeJam & Earl are the cream of the co-op crop in my opinion. You can't go wrong with any of those.
i second sunset riders, mickey and donald, alien storm (very underrated!!) and two crude dudes - which all tip top titles....
if you want something thats not a brawler / platformer - get columns!! great game, in particular in cooperative two player mode :)

I don't know Virocop at all, I'll have to check into that too. Gunner Pilot stuff always failed horribly for me in Shadow Squadron and Star Wars Arcade, but mainly because the pilot's movement affected the crosshair. I guess that isn't a problem in Soul Star (not that I will ever experience this first hand)?
I don't know if I'd call it the best, but probably the most memorable for me was Toejam & Earl. I thought that game was hilarious and infinitely re-playable. Me and my buddies would play so much of that game while just fucking around. The random levels really helps.
(Edit, I just had an impression that I might be mistaken about player 2 controlling shield/engine power. I'll have to try to confirm that. My memory might be getting confused.)
[The co-pilot is controlling weapons and shield/engine power.
Teamwork and communication is required. As the pilot I could call for my engine power transfered to shields in hairy situations, and there was also communication about what weapons to use. It added quite well to the intensity, and surprisingly my friend really loved playing as weapon/tactical/co-pilot. More surprisingly, we even beat the game in two player mode.]
How do you hold those joysticks? Do you fasten them down or something? I know there was an Atari joystick coupler, but it's pretty rare.
Sonic 2 (as well as Sonic 3, Sonic 3 & K) are a lot of fun to play in co op.
That's interesting that you were able to do that, but I can not imagine anyway possible to make that work easily for Sega-CD. The multiple weapons are mapped to different buttons. You must have quite the dexterity. And I don't see how one button would cut it.
The real was to do it is to play with another human being. You know, co-op.

Ha! Good one. I knew Soul Star is (unfinished) on Jaguar CD, and it was planned for 32x, so it wasn't a stretch to figure maybe there was a CD32 release I'd somehow not known about.
And anyway, the posts SonicRetard quoted and responded to were all referring to Soul Star.

I love Battletech but not the co-op mode. Same goes for LHX. Both are usually an exercise in frustration when played with a friend. Though not as bad, games like Sonic 2 also can be frustrating in co-op; the player that gets Tails gets left behind too much. If 32X games count, then include Knuckles Chaotix to my list of favorites because it's great with two players, even better than the one player mode.
