Definately not the NES.
I would go with NES, though the absolute best are probably 16-bit (Secret of Mana, Hard Corps, Putty Squad, Streets of Rage 2, the Genesis version of Lost Vikings, Turtles in Time, Gunstar Heroes, Pocky and Rocky), there's just so much co-op goodness on the NES. I tend to think that the small sprites and zoomed out graphics of 8-bit suit co-op better as well to be honest.

Battletoads ? Come on man, everyone knows its a horrible 2 player game and shouldnt even be 2 player.
Very close all round. The Genesis for GA and SoR both 1 and 2 plus a few shooters and World of Illusion; CHC being too tough for most player pairs. SNES has DKC 1 and 2, TMNT in Time, and numerous brawlers of a mediocrity that matches the Genesis's co-op shooters'. The NES has TMNT 2 and 3, the best Double Dragons and Contras, along with Mario 3 (not simul co-op but with each player clearing levels it feels cooperative despite the old arcade battles that arise) and a far better lightgun selection.
So NES.
Easily the Megadrive/Genesis for me. Even just for the Streets Of Rage and Golden Axe games alone.
Genesis for the multitude of beat em ups and sports titles. I can remember playing NHL hockey with 3 other friends using the multi-tap. The SNES and NES both have their shares of co-op games, but I tend to think of the NES and SNES more as one-player machines. NES is most notable for its platformers and the SNES most notable for its RPGs. I'm simplifying of course, but this poll is based on broad generalizations anyway.
The Genesis/Mega Drive, and for many of the same reasons people have already stated it above. Games like Streets or Rage, Golden Axe, Gunstar Heroes, and many more. The Genesis is great for co-op games.

Not necessarily, there was a thread on what console had the "Best RPG's", and SNES had the most votes. There was also a thread on "NES vs SMS", and NES got the most votes, so there's really no bias here. The Genesis had lots of great Co-op games, and my friends and I had a lot of fun playing them back in the day, and we still do now.

Altered Beast, Golden Axe, Golden Axe II, Alien Storm, Two Crude Dudes, Streets of Rage, Streets of Rage 2, Streets of Rage 3, Double Dragon, Double Dragon II, TMNT Hyperstone Heist, The Punisher, Captain America and the Avengers, World of Illusion, Ren 'n Stimpy, Toejam & Earl Panic on Funkatron, Contra Hardcorp, Gunstar Heroes, Rolling Thunder 2, Bonanza Bros., Toejam & Earl, Crack Down and Skeleton Krew, make the Genesis the best coop platform possibly of all time.
Add the 32X and Shadow Squadron is great fun as well. Add the Sega CD and Final Fight CD is a terrific addition.
On the NES I remember playing Life Force, which really has no equal in my opinion. Scramble Spirits on Master System and Vapor Trail on Genesis just don't fill that void for me. TMNT the Arcade game was amusing, and I know people swear by the third game but I was never impressed by it. I guess the Contra games on NES are great, I'm pretty sure Double Dragon II is coop. That is literally all I remember playing at friend's houses back in the day, and all of my friends owned an NES.
The Super Nintendo doesn't even compare to the NES in coop games, much less the Genesis.
Aren't there any Co-Op games of note for the Sega Master System?
