Considering it was a Disney (American) joint operation with Virgin (British), I'd opt for Disney's Aladdin. However, Vectorman is right at the top of my list as well.
I think I would have to agree with both of you. Vectorman and Disney games.
I disagree with that, but hey, you like Goofy's Hysterical History Tour and I don't. ;)
My favorite American-made game? Off the top of my head, Eternal Champions, but I'm going to disagree with that most likely since there are faaaaaar too many games to choose from.
Wing Commander and Rise of the Dragon.
Not knowing Blue Sky was an American company, my first response was, "Is this a trick question?" The lion's share of good Genesis games come from Japan. However, here are some of my American-derived faves:
Vectorman
Comix Zone (formerly Aladdin:))
Lost Vikings
Rock'n Roll Racing
Hardball 94

I think Aladdin is certainly a bit of an iffy choice for the thread seeing as it was both designed and programmed by brits, but I guess the US musician and artwork people bring it to a kinda' 50/50 game between the countries.
Some other games that I just remembered that deserve merit from the US have to be
Starflight
General Chaos (think that was US made)
Desert Strike
Toejam and Earl games
Comic Zone
Dune II
The Marvel Comics games (Spiderman, X-men)
Lion King (that one was made in the US totally I think)
I'm with Baloo on this one. Eternal Champions immediately popped into my head.
Vectorman, X-men 2, and Earthworm Jim.

Gotta be Dune II. I just can't stop playing that damn game.
Somehow I never realized that it's an American game. I always assumed that Westwood Studios would be based in the UK.
Anyway, the game's great, and the Mega Drive variant is as good if not better than the MS-DOS original.
