As tempted as I am to say Sonic the Hedgehog, I can't think of a more over rated Genesis game than Vectorman.
I haven't played all the way through it, so I can't fully judge it as an overall game. But the graphics are certainly over rated enough to make up for anything else good about it and all I ever here about it is talk about the graphics anyway.
Altered Beast is up there. I don't know how popular it would've been if people had paid full price for it. Its still cool for what it is.
I'd say Strider, since I've never liked it and think the gameplay is bad. But the Genesis version is coasting on the arcade original's over-ratedness.

But on one hand, this is highly subjective as not everyone will understand the praise certain titles get. I personally disagree with the sentiments here about MUSHA and Gleylancer. The latter of the two got it's notoriety through its rarity combined with whatever quality was within. The same thing could have happened with Dangerous Seed, which was also quite nice. . .but not nearly as rare. Any rare game that happens to be of some quality will get
These days, just about everything gets praise so the media sources can keep whatever ties to publishers that allow for 'exclusive' features and interviews. With so many people reminiscing about the past, it's easy for some to enjoy one title greatly while others that investigate don't find anything that amazes them.
So I'm just going to go out and say that every single game ever created can justifiably be labeled as over-rated.

BTW, just to divert the topic a bit (it IS Sega-16 after all), who agrees that Halo: Combat Evolved is the most overrated game of all time?
I never actually played the first Halo until I bought it a few days ago, and I agree its overrated - I've been playing PC games like Halo, years before it came out.
I still think it's a decent game.
